Abstract   In this study, the concentration of tumorassociated antigens, p97 and p250, were determined by autoradiographic method in tissues of malignant melanoma. We tried to establish in vitro guantitative autoradiography, and found that 20 micron frozen sections incubated 1 hour at room temperature with varying concentrations of I-125 labeled monoclonal antibodies were suitable for this purpose. Digitized autoradiographic images from these sections were used to calculate the maximal binding values(Bmax) which were equal to the concentrations of antigen or epitope. In the experiment of cell block, the Bmax obtained from this method was found to be close to the concentration of antigen calculated from Scatchard analysis. P97 concentration could be measured in 31 specimens out of 36 cases of malignant melanoma, and 74% of tumors expressed p97 antigen. P250 concentration was measured in 33 specimens, and 26 out of 33 samples(79%) had detectable antigen expression. P97 concentration ranged from 14.4 pmoles/gram to 368.8 pmoles/gram with a median value of 40.6 pmoles/gram. P250 antigen ranged from 6.0 pmoles/gram to 431.1 pmoles/gram with a median value of 62.0 pmoles /gram. Autoradiographic results correlated well with gradings of immunoperoxidase staining. In case of p 97, the correlation coefficient was O.806(p<0.001) and in case of p 250, 0.875(p< 0.001).
